Ever since The Joker was announced to be in David Ayer's Suicide Squad many speculated what exactly his role will be in the film. Many for some reason thought that he was the movie's antagonist that the Suicide Squad and love interest Harley Quinn will have to stop. Well this seems to be incorrect.
According to Latino-Review The Joker played by Jared Leto will play a 'Hannibal Lector' type role throughout the film up until the end. Harley Quinn will try to bust him out of Arkham towards the beginning until Amanda Waller moves him to Belle Reve. This type of role is played out in Batman: Assault on Arkham animated film WB released last year.
The real enemy for Task Force X seems to be Jesse Eisenberg's Lex Luthor. In the film he seems to be the one who has illegally obtained bio weaponry and mutated species including King Shark. It's still unknown if Eisenberg will have time to film this movie but it's entirely possible to simple be 'behind the scenes' on all the mayhem that will transpire in the film not needing Eisenberg to physically appear in the movie.
Lastlym they revealed that Rick Flagg (Once rumored to be Tom Hardy) is sort of the good guy of the squad keeping everyone in check including Amanda Waller. They reveal this film will lay the ground work for multiple other DC franchises for the not to distant future.
